Efficacy and Safety of the Dual SGLT1 and SGLT2 Inhibitor JP-2266 in Type 2 Diabetes Mellitus: A Randomized, Double-Blind, Placebo-Controlled, Phase 2 Study.

Dual SGLT1/SGLT2 Inhibitor JP-2266 Significantly Reduces HbA1c in Type 2 Diabetes Phase 2 Trial — Both doses of JP-2266 produced clinically meaningful, placebo-adjusted reductions in HbA1c of approximately 0.94–0.97 percentage points over 12 weeks in patients with type 2 diabetes inadequately controlled by lifestyle modification alone (5 mg: −0.94%, 95% CI −1.24 to −0.65; 10 mg: −0.97%, 95% CI −1.26 to −0.68; both P<0.0001).

Impact of Paclitaxel Coated Devices on Limb Salvage and Target Vessel Revascularisation Rates following Isolated Femoropopliteal or Infrapopliteal Endovascular Revascularisation for Chronic Limb Threatening Ischaemia: Insights from the SWEDEPAD 1 Trial.

Paclitaxel-Coated Devices Show No Limb Salvage Benefit in Femoropopliteal or Infrapopliteal CLTI — In a prespecified subgroup analysis of the SWEDEPAD 1 trial involving 1,778 patients, paclitaxel-coated devices failed to improve limb salvage over uncoated devices in either the femoropopliteal or infrapopliteal segment, and an apparent one-year reduction in target vessel re-intervention (TVR) in the femoropopliteal group — about 27% lower risk (HR 0.73, 95% CI 0.56–0.96) — was not confirmed by interaction testing or composite endpoint analysis.

Continuous or Fixed-Duration Maintenance Therapy in Multiple Myeloma.

Fixed Two-Year Lenalidomide Maintenance Matches Indefinite Therapy in Overall Survival — In a phase 3 randomised trial of 516 patients with standard-risk newly diagnosed multiple myeloma, indefinite-duration lenalidomide maintenance produced virtually identical seven-year overall survival compared with a fixed two-year course — 68.6% versus 69.0%, a difference of just −0.4 percentage points (95% CI −9.0 to 8.3; P = 0.93).

Baloxavir marboxil, favipiravir, or oseltamivir in patients with non-severe symptomatic seasonal influenza (AD ASTRA): a phase 2, open-label, adaptive, randomised controlled trial.

Baloxavir Shows Greatest Viral Clearance Among Three Oral Influenza Antivirals in Head-to-Head Trial — In a randomised controlled trial of 944 adults with early seasonal influenza, baloxavir accelerated oropharyngeal viral RNA clearance by 86% compared with no treatment (95% CrI 60–117), outperforming both favipiravir at 66% (95% CrI 45–94) and oseltamivir at 49% (95% CrI 28–74), though none of the three agents meaningfully shortened the time to complete symptom resolution.

Extended Dual Antiplatelet Therapy for Multivessel Coronary Artery Disease.

Extended DAPT Reduces Cardiovascular Events in Multivessel CAD Without Increasing Bleeding — In a large randomized trial of 8,250 patients with multivessel coronary artery disease who remained event-free after 12 months of standard dual antiplatelet therapy, extending treatment with clopidogrel plus aspirin for an additional year reduced the composite risk of cardiovascular death, myocardial infarction, or stroke compared to aspirin alone — roughly an 18% lower relative risk (HR 0.82, 95% CI 0.69–0.98; p=0.03).

Targeting depression before it starts: An updated systematic review and meta-analysis of preventive interventions in young adults from randomized controlled trials.

Psychosocial Interventions Modestly Reduce Depression Symptoms in Young Adults, But Effects Fade — Across 65 interventions from 58 randomised trials, psychosocial preventive programs produced a modest reduction in depressive symptoms in young adults at the end of treatment, equivalent to a small-to-medium effect (Hedges' g = −0.52, 95% CI: −0.70 to −0.33), with a possible reduction in the risk of developing major depressive disorder of approximately 23% (RR = 0.77, 95% CI: 0.61 to 0.97).

Optimizing prostate biopsy decision making – (MUSIC-screen): A 1:1 randomized controlled trial comparing micro-ultrasound versus multiparametric magnetic resonance imaging for prostate cancer diagnosis.

Phase 3 RCT Launched to Test Whether Micro-Ultrasound Matches MRI in Prostate Cancer Screening Decisions — MUSIC-Screen is an ongoing phase 3 noninferiority randomised controlled trial enrolling 1,284 men to determine whether micro-ultrasound (microUS) is non-inferior to multiparametric MRI (mpMRI) for guiding prostate biopsy decisions and detecting Grade Group ≥2 prostate cancer; no outcome data are yet available.

Facilitating Communication for Family Members of Critically Ill Patients During and Following a Hospital Stay: A Randomized Controlled Trial.

Nurse Communication Facilitator Intervention Fails to Reduce Depression in ICU Families — A multicenter randomized controlled trial found that involving trained nurse communication facilitators during and after an ICU stay produced no meaningful reduction in depressive symptoms among family members of critically ill patients, with a negligible and statistically non-significant difference in mean depression scores between groups over six months (difference in mean HADS-depression: −0.07, 95% CI −0.70 to 0.55).

SGLT2 Inhibitor on Infarct Size and LV Remodeling by CMR in Patients With AMI.

Empagliflozin Shows No Reduction in Infarct Size or LV Remodeling After AMI — In a randomized controlled trial of 200 high-risk AMI patients, empagliflozin 10 mg daily did not reduce infarct size compared with control at 6 months (12.5% vs 12.9% of LV mass; no statistically significant difference, P = 0.92), nor did it promote reverse left ventricular remodeling (change in LV end-systolic volume: −3.3% vs −6.8%; P = 0.40).
